Abstract
ESCALONA V., Jesús Alejandro; LAYTON T., Cristian Fabián and MENDIETA Z., Hugo. Standardization of the mRNA extraction method from umbilical cord vessel. Rev. mex. cienc. farm [online]. 2015, vol.46, n.2, pp.50-53. ISSN 1870-0195.
Gene quantification in umbilical cord has been used to study several diseases, however the correct homogenization of the vascular tissue is a latent problem. The main objective of this study was to propose a sequence of steps leading to the successful mRNA extraction from umbilical cord vessels. 20 samples of umbilical cord, from which the vessels were dissected to be homogenized in the Bullet Blender Standard BBX24 (Next Advance, Inc., Burden Lake Road, NY, USA) were processed. mRNA extraction was performed on the MagNA Pure LC 2.0 (Roche, Germany), using the MagNA Pure LC kit RNA Isolation Kit III (Tissue) (Roche, Germany) and its concentration was quantified by spectrophotometry at 260 nm in a NanoPhotometer (Implen GmbH, Germany quantified ). mRNA was obtained in 100% of the samples.
