In the present study we observed the effect of nimesulide with the objective of evaluating their effect in osteoclast formation. We used Sixty male Wistar rats weighing around 250 ± 25 g. A uniform standardized U spring, made of 0.012-inch nickel-titanium wire was set in each animals mouth between the right and left upper first molar. The animals were divided into 4 groups and three of them were prescribed with low, medium and high doses of nimesulid. The animals were killed at 24, 48 and 72 hours after the placement of the appliances. The maxillae was removed and fixed in formaldehyde for 48 hours, and subsequently decalcified in EDTA (pH 7.4), and included in Paraplast. Serial sections of 7 micrometers were cut and stained with hematoxylin and eosin. Results: the NSAIDs nimesulide in all doses inhibited the formation of osteoclasts. At 24 h the osteoclast inhibition was dosis dependent. At 48 and 72 h all doses were effective. In control group, osteoclasts increase gradually at 24, 48 and 72 h.
