The surface plasmon resonance (SPR) finds a directly application like an optical sensor during biomolecular interaction reading. In this work we applied a surface plasmon resonance microscope (SPRM) to simultaneously and in real time detect the protein-protein interactions combined with microarrays. The detection of those biomolecular binding events was carried out label free. The fabricated microarrays contain 144 binding active microsites of protein from (1) human serum albumin HSA (0.1, 05, 1.0 mg/ml), (2) lyzosime Lyz (0.1, 05, 1.0 mg/ml), (3) anti bovine serum albumin anti BSA (1.0 mg/ml), and (4) bovine serum albumin BSA (0.1, 05, 1.0 mg/ml). We determined simultaneously the surface density concentration when anti BSA (1.0 mg/ml) was injected onto the microsites of protein by using the SPR kinetic interaction curves. We found that the surface density concentration of anti BSA (1.0 g/ml) during the interaction with active microsites was 660 ng/cm2 for BSA, 366 ng/cm2 in the case of HSA, 22.3 ng/cm2 with Lyz, and 8.3 ng/cm2 for anti BSA.
