Many cellular processes are regulated by reversible protein phosphorylation. cAMP dependent protein kinase (PKA) is present in all eukaryotes (protozoa, fungi, algae, and animals) with the exception of land plants and its study has revealed important mechanisms for cell signaling. Here, the components of the PKA signaling cascade in Saccharomyces cerevisiae, the compartmentalization of the regulatory subunits as well as the mechanisms for regulation of the stress response, the stimulation of filamentous growth, and the feed-back regulation of PKA activity are described. The genetic analyses of the components of the PKA pathway in S. cerevisiae have revealed detailed information about its evolution, structure-function relationships, and operation by means of mathematical and computational modeling.
